The Relationship Between Type 2 Diabetes and Osteoporosis

Researchers conducted a comprehensive study, collecting data from 490,089 patients with Type 2 Diabetes Mellitus (T2DM), also known as type 2 diabetes, obtained from the Genome-Wide Association Study (GWAS) database. In addition, they also used osteoporosis data from two large sources, namely FinnGen (212,778 patients) and MRC-IEU (463,010 patients).

For the causal analysis, the researchers explored the role of three obesity-related characteristics (BMI, waist circumference, and waist-to-hip ratio) as well as 30 mediating variables related to metabolism and inflammation.

Research Results

Causal Relationship

This study found a strong causal relationship between type 2 diabetes and osteoporosis. Patients with type 2 diabetes have a higher risk of developing osteoporosis, with an odds ratio (OR) greater than 1.5 and a p-value of less than 0.001. Steiger test results also showed no reverse causality between the two conditions.

Role of Metabolic Factors

Although the relationship between type 2 diabetes and osteoporosis is strong, after adjusting for factors such as BMI, waist circumference, and waist-to-hip ratio, the direction of the relationship remained consistent. However, no specific risk factors related to glycolipid metabolism, amino acids, or inflammation were found to mediate this causal relationship.

Clinical Implications

This study underscores the importance of managing type 2 diabetes by paying attention to metabolic factors such as BMI, waist circumference, and waist-to-hip ratio in order to prevent complications such as osteoporosis. Although no specific metabolism-related mediators were found, efforts to control other metabolic factors remain important in the management of type 2 diabetes and the prevention of osteoporosis.
